Meclofenoxate
Drug form and composition
Powder for solution for injection and solvent. One ampoule of 10 ml contains
250 mg Meclofenoxate hydrochloride (powder for solution for injection). One ampoule
of 10 ml contains 10 milliliters Sterilised water for injections (solvent).
Indications
The preparation is indicated in following conditions: comma; skull and brain
trauma; following a stroke; encephalopathy; mental disorders (in combination with
psychotropic agents); mental and psychomotor retardation in children; brain intoxication;
alcohol psychoses; neuritis and polyneuritis.
Dosage and administration
The preparation is administered intravenously, subcutaneously and intramuscularly,
most frequently as a single dose of 250 mg (1 ampoule). The powder is diluted just
before injection. Depending on the patient condition the dose may reach 1-2 g/24h.
Duration of the treatment depends on the severity of the disease and should be determined
by the doctor.
Contraindications
Hypersensitivity to the preparation; infection of the CNS; epilepsy; neuroses
presenting with intension and phobia; renal and hepatal insufficiency.
Special warnings and precautions
In patients with severe arteriosclerosis and in elderly patients the preparation
should be used with care, as a risk of significant agitation exists. The medicine
should be administered in pregnant and nursing women only if serious indication
exist and on doctor’s decision.
Drug interactions
Centrophenoxin is used as a basic therapy in mental disorders. In such cases
it is combined with psychotropic agents (antipsychotics, anxiolytics, and antidepressants).
In the clinic practice Centrophenoxin is successfully combined with antiepileptic
and anti-Parkinson’s disease medicines. In cases with alcohol and CNS - depressing
agents intoxication Centrophenoxin exerts a detoxication effect. In concomitant
treatment with psychoanaleptics an increase of CNS excitation may appear.
Adverse reactions
In isolated cases in predisposed patients, particularly in elderly persons, the
preparation may provoke excitation, aggression, insomnia, and headache. In patients
with paranoid symptoms and hallucinations the preparation may lead to worsening
of the condition. In some cases skin allergy and gastrointestinal disorders may
appear.
Pharmacological mechanisms
Centrophenoxin belongs to the group of psychostimulants - nootropic agents, which
possess an activating action on the brain, improving the neural cell metabolism.
Centrophenoxin stimulates the aerobic glycolysis and leads to improvement of the
glucose utilization in the brain cells. It improves the brain cell stability to
hypoxia and toxic agents or their metabolites. In long-time use the preparation
recovers the functionally injured brain cells.
Supplied
5 ampoules of 10 ml (250 mg) containing powder for solution for injection, and
5 ampoules of 10 ml containing solvent.
