- COMPOSITION:
Progesterone 0.005 mg/ 1 ml; 0.01 mg/1 ml in injection solution.
ACTION:
Natural hormone of corpus luteum, synthesized and excreted by the
ovaries, adrenals and during pregnancy - by the placenta. Its basic
biological action is exerted on the sexual organs and mammary
glands. Its specific effect is expressed in preparation of the
uterine mucosa to receive and fix the fertilized ovum, and for
development of the fetus. It reduces the excitability and
contractility of the uterine and tube muscles, stimulates the
proliferation of the glandular epithelium and the acinous part of
mammary glands, preparing them for lactation. The elevated
progesteron level inhibits the release of luteinizing hormone from
the pituitary gland.
PHARMACOKINETICS:
Progestron (and its derivatives) are metabolized in the liver by
hydroxylation and by reduction of the ketone group, and are excreted
with the urine after being transformed into non-active metabolites.
INDICATIONS:
Habitual abortions or proneness to such abortions; imminent
abortions; dysfunctional uterine hemorrhages; uterine hypoplasia and
amenorrhea; dysmenorhea; infertility by anovular cycle.
CONTRAINDICATIONS:
Progestron is not recommended to be prescribed during menstruation
and after the 37th week of pregnancy. Hepatic and renal damage. It
is not recommended in nervous and psychic diseases. |
SIDE EFFECTS:
Nausea, vomiting, diarrhea, fluid retention, headache, reduced
libido. Sometimes edemas and albuminuria with hypertension can occur
when administered during the first months of pregnancy.
DRUG INTERCATIONS:
The effect of progesteron is reduced by amplicillin, barbiturates,
carbamazepin, phenytoin, rifampicin. The effects of sulphanylurea
antidiabetic agents are diminished by simultaneous administration.
Progestron diminishes the action of the uterotonic drugs, anabolic
steroids and gonadotropic hormones from the anterior lobe of the
pituitary gland.
DOSAGE AND ADMINISTRATION:
It is injected intramuscularly according to an individual schedule
for each patient, according to the indications for progesteron
treatment. The doses in habitual abortion (0.005-0.01 g, 2-3 times
weekly) are lower than those in cases of imminent abortion
(0.01-0.25 g every day). Progesteron treatment of sterility and
hormonal dysmenorrhea is preceded by the administration of
follicular hormone - during the first phase of the menstrual cycle.
The same schedule is applied also in hypoplasia of the uterus,
amenorrhea and dysmenorrhea.
