- COMPOSITION:
Coffeine-sodium benzonate 100 mg/1 ml, 200 mg/l and 250 mg/l in
one ampoule.
ACTION:
Stimulator of CNS, an analeptic drug.
Therapeutic doses enhance and regulate stimulation processes in the
brain cortex, enhance the positive conditioned reflexes and increase
the motor activity, dectreasing the fatigue and drowsiness. Coffeine
is a direct antagonist of alcohol and of narcotic drugs. It
potentiates cardiac contractions, dilates the cerebral vessels, the
coronary vessels, the vessels of the skeletal muscles and the lungs.
Under the influence of coffeine in collapse and shock conditions,
the blood pressure increases. It enhances the diuresis and increases
the metabolism. It increases the intracellular comulation of AMP
through phosphodiesterase inhibition, and this effect explains the
enhancing action of glycogenolysis and metabolic processes in
different organs.
PHARMACOKINETICS:
Coffeine is a very weak base which is quickly absorbed and it
stimulating effect is manifested 5-8 min after subcutaneous
application. It is uniformly distributed in the different tissues of
the organism and passes through the blood-encephalic barrier and
placenta. Its plasma half-life is about 4h. it metabolizes fully in
the liver by demethylation and is excreted with the urine in the
form of metabolites in 90-92% and in 8-10% - unchanged. Slightly
excreted through skin as well.
INDICATIONS:
Coffeine-sodium benzoate is administered as an analeptic drug in
infectious diseases, intoxication with narcotics and other
substances, especially when the inhibition of the centers in medulla
oblongata is accompanied by disturbances of consciousness. In cases
of acute vascular insufficiency and acute cardiac failure, also in
chronic cardiac insufficiency, especially with bradycardia. It is
used to improve the physical and psychic working capacity. In case
of headache and migraine (combined with analgesics). |
CONTRAINDICATIONS:
Increased excitability of CNS, insomnia, peptic ulcer, glaucoma;
heavy forms of hypertension and pronounced atherosclerotic vascular
changes. The preparation should be applied carefully in cases of
tachycardia and arrhytmia.
SIDE EFFECTS:
Insomnia, nervous excitement, light tremor; in high doses -
headache, hallucinations, dizziness, accelerated pulse and
respiration, vomiting and diarrhea, possibly convulsions.
DRUG INTERACTIONS:
The half-elimination of theophylline and that of coffeine are
prolonged in newborn and prematurelly born babies, and additive side
effects may be observed.
DOSAGE AND ADMINISTRATION:
Parenterally (usually subcutaneously, rarely intramuscularly and
intravenously) - 1 ml (i.e. 0.1, 0.2) or 0.25.
MEDICINAL FORM AND PACKAGE:
Ampoules of 100 mg - 1 ml (10 %), 200 mg - 1 ml (20%) and 250 mg - 1
ml (25%) in packs of 10 and 50.
STORAGE:
At moderate temperature (15-30 C).
EXPIRY:
3 (three) years. |
